起源NOD-SCID小鼠是由非肥胖性糖尿病小鼠(NOD/Lt)和SCID小鼠杂交建立的一种重度免疫缺陷小鼠。相对于SCID小鼠,NOD-SCID小鼠T、B淋巴细胞缺陷,各种肿瘤细胞可以植入,且较少发生排斥反应及移植物抗宿主病(graft-versus-host, GVHD)。
NOD-SCID的小鼠还具有NK细胞活性低、无循环补体、巨噬细胞和抗原呈递细胞(antigen-presenting cell, APC)功能受损的特点。这种重度免疫缺陷小鼠的缺点是仍有残留的NK细胞活性,生存时间比SCID更短,限制了长期研究。NOD-SCID小鼠因易产生胸腺淋巴瘤,也是导致其寿命较短的因素之一。
特征与用途
◇病理:非肥胖糖尿病重症联合免疫缺陷。
◇用途:常用于肿瘤、免疫学、胚胎干细胞、艾滋病及肝炎研究,为T' B' NK细胞缺陷的严重联合免疫缺陷动物模型。人造血干细胞领域、肿瘤细胞等移植方面的研究。通过对NOD SCID进行细胞移植来研究糖尿病发病原理。
目前国际公认的免疫缺陷程度最高、最适合人源细胞移植的工具小鼠,由于不同开发公司命名不同,NOG和NSG、NPG、NDG的缺陷基因是一致的,均为NOD- Prkdcscid -IL2rgtm1突变纯合子小鼠。
