   本课题组主要研究方向是在上皮-间质细胞转化(Epithelial-mesenchymal transition, EMT)和肿瘤转移中基因网络的表达与调控。具体研究对象是转录因子Snail抑制基因转录的分子机理。Snail属于含有SNAG结构域的锌指(zinc-finger)蛋白家族成员。SNAG位于Snail分子的氨基端,具有抑制基因转录的功能。在胚胎发育和肿瘤转移过程中,Snail是诱导EMT的关键因子,是连接多种促肿瘤转移信号通路的中心分子。因此,研究Snail在细胞内的作用机理不仅在理论上有助我们弄清楚肿瘤转移的机制,在临床上也能够提供新的靶点进行药物设计和制定有效的治疗方案。

   当前正在对组蛋白精氨酸甲基化在肿瘤转移中的作用进行探讨。组蛋白精氨酸残基甲基化对基因的表达与调控具有重要作用。但是相对于其它组蛋白修饰例如磷酸化,赖氨酸的甲基化等,精氨酸的甲基化研究没有因以足够的重视。精氨酸甲基转移酶VPrmt5)属于II型精氨酸甲基转移酶,除了能够甲基化修饰组蛋白H2AH3H4上的精氨酸残基,还能够修饰非主蛋白底物例如转录延长因子SPT5Prmt5 通过甲基化修饰组蛋白来直接负调控基因表达,也能够用过甲基化修饰来调控蛋白复合体的组装来间接调控基因表达。我们前期研究发现了Snail同过结合AjubaPrmt5来抑制转录,其中Ajuba作为支架蛋白,连接SnailPrmt5共同形成一个多蛋白复合体。

2003年于美国辛辛那提大学医学院获生理学博士学位,宾夕法尼亚大学博士后, Wistar研究所任研究助理(staff scientist),期间曾获美国国防部Idea Award 专项基金支持。

20112月回国,任上海交通大学医学院生化与细胞系研究员,基因表达与调控课题组长。主要研究转录因子在肿瘤发生和转移的基因网络的表达调控,论文发表在PNASCancer ResearchNucleic Acid Research等刊物。
