
Labelled quantitative analysis ITRAQ/TMT
For protein samples that require labeled quantitation, after extracting high-quality proteins, isotopic labeling is required for the samples. The labeling reagents can be selected based on the number of samples (iTRAQ: 2-8 samples, TMT: 2-10 samples).
Label-free quantification
For label-free quantitation (LFQ) analysis in proteomics, there are two main methods: DDA/DIA (SWATH). Label-free quantitation methods do not require labeled reagents, have minimal interference with the system, and have relatively low costs. There is also no limit to the number of samples that can be quantified simultaneously, ranging from 2 to an infinite number of states. The differential analysis mainly relies on software parallel comparison and calculation, so the software requirements are relatively high. In the case of a small number of samples, we recommend the DDA-LFQ method, and in the case of a large number of samples that need to be quantitatively screened, we recommend the DIA (SWATH)-LFQ method. The latter has a more complex experimental process and data analysis, but can parallel detect dozens to hundreds of biological samples.
