Lab Info:Prof. Hao Wang’s research group relies on pharmacy discipline of Shanghai Jiaotong University School of Medicine, the construction discipline of national "double first-class" and "high-level local universities" in Shanghai. At present, there are one Professor, two associate professors, one experimenter, one postdoctoral and five graduate students in the group. Over the past twenty years, the research group has focused on the new pathogenesis of neurodegenerative diseases, new targets for intervention and new drug research and development. It has successively undertaken one National Key Basic Research Program of China (973 Plan), one National Science and Technology Major Project of "new drug creation and development", twelve National Natural Science Foundation Projects and three key projects of Shanghai Science and Technology Commission. It has published more than 60 papers in high-level magazines such as Cell Death & Differentiation, Advanced Science and Aging Cell, and has obtained more than 10 invention patents. The achievements won the first prize of the science and technology progress award of the Ministry of education, the second prize of the natural science award of the Ministry of Education, the second prize of the Chinese medical science and technology award, etc. Professor Wang Hao, the Principal Investigator, has served as the director of Chinese society of Pharmacology, director of Shanghai Pharmaceutical Association and vice chairman of the special committee of pharmacology, won the woman pace-setter of Shanghai education system, Shangyao Xinglin talent cultivation award, and trained five doctoral students and eight master's students, and four of them won national scholarships.

Lab Info:Drug resistance and tumor relapse team mainly uses systems biology approaches (integrated genomics, cellular and molecular biology, biochemistry and animal models) to study the molecular mechanism of pediatric leukemia drug resistance and tumor relapse for both basic and translational intents. The team currently consists of 1 CO-PI, 5 research technicians, 2 postdoctoral fellows and 5 PhD candidates. Three previous postdoctoral fellows had been appointed as associate and assistant professor respectively. Three PhD graduates are engaged in post-doctoral trainings in the United States and China respectively.

Main research areas:

1. Basic mechanism and translational application of drug-resistant and tumor relapse in childhood leukemia.

2. Role of DNA damage response (DDR) in drug-resistant and tumor relapse of childhood leukemia.

Mental disorders are diseases caused by dysfunctions of the brain's neural circuitry, leading to varying degrees of impairment in cognition, emotion, behavior, and volition. These include conditions such as schizophrenia, depression, and autism spectrum disorder. It is estimated that approximately one billion people worldwide are affected by mental disorders, with a trend of increasing prevalence year by year. The causes of mental disorders are highly complex and are generally believed to be associated with disruptions in the neurotransmitter systems of the brain. As a result, neurotransmitter-related proteins have become major drug targets for the treatment of mental disorders. Ion channels play a critical role in neurotransmitter-mediated neuronal signaling. Research into their regulatory mechanisms and pharmacology helps elucidate the underlying pathology of these diseases and guides drug development. Our research group focuses on drug targets related to neuropsychiatric disorders. By integrating interdisciplinary approaches such as structural biology, pharmacology, and computational biology, we investigate the mechanisms of novel targets and conduct research on the design and discovery of innovative drugs. The main research directions include:

1)Regulatory mechanisms of ion channel proteins associated with neuropsychiatric disorders

2)Pharmacological mechanisms of drugs used to treat neuropsychiatric disorders

Lab Info:Her group has been focusing on the creation of natural and biomimetic nanocarriers for achieving efficient and precise brain-targeted delivery of biomacromolecule medicines. The major work include: the development of lipoprotein biomimetic nanocarrier with multi-mode drug loading capacity and tunable targeting property; the disclosure of the pathway and mechanism for brain clearance of nanocarriers; and the applications of nanotherapeutics for the management of brain diseases. Her group has published over 60 articles in high-impact journals such as Nature Communications, Adv Science, ACS Nano, Nano Lett, and J Control Release, among others. Their work has been acknowledged and featured in Sci Transl Med, ACS Nano, and other journals, as well as in well-known professional media outlets. Her group had applied and gotten approved more than 20 innovation patents.

Lab Info:The human brain is made up of billions of neurons, which in turn communicate information with other neurons through trillions of synapses. Synapses are the basic structure of neuronal communication, synaptic plasticity and its precise regulation play an important role in various brain activities, including cognition, perception, decision making and sleep/awake balances. Whereas synaptic abnormalities or disorders are often accompanied by the occurrence of various brain diseases, such as Alzheimer's disease, autism, depression, etc. At present, our research group mainly focuses on the synaptic regulation to understand the pathogenesis of neurodegenerative diseases and psychiatric disorders. Research is carried out mainly from the following three directions:

1) Develop and optimize new tools for brain science research, namely new genetically encoded neurotransmitter fluorescent probes;

2) Explore the regulatory mechanism of synaptic transmission with multi-channel whole-cell patch clamp recording technique;

3) Understand the molecular and cellular mechanisms of synaptic regulation in Alzheimer's disease (AD) and autism spectrum disorders (ASDs).

Lab Info:The research field of my lab spans a broad range of chemical biology, structure biology, epigenetics and drug discovery. We are interested in studying the dynamic catalytic and regulation mechanisms of chemical modifications on biological macromolecules involved in Fatty acid biosynthesis, Nucleoside biosynthesis, and DNA/RNA methylation/demethylation pathways. These processes are vital for all living cells, and the biofunctions of enzymes involved in these processes are restrictively regulated spatio-temporally through chemical modifications. Our researches focus on elucidating the mechanisms of the key enzymes in adding, reading and removing the chemical modifications on the biological macromolecules, as well as the subsequent corresponding biological effects. We are also establishing efficient high throughput drug screening systems for novel leading drug compounds discovery against these potential drug targets. So far, our lab has published several research articles on Nature Chemical Biology, Nature Communications, Cell Research, PNAS, Chemical Science, Elife, etc, and obtained 2 national patents and applied a domestic patent.  Our works are supported by numerous grants such as the National Science Fund for Excellent Young Scholars, the Major Research Plan and General Program Research Plan of National Natural Science Foundation of China, Shanghai Shuguang Scholar program, Oriental Scholar of Shanghai College, and Shanghai Biomedical science and technology innovation program, etc.

Lab Info:Our group has been dedicated to develop visualized drug delivery systems with high sensitivity and high selectivity for disease diagnosis and therapy, by using interdisciplinary technologies and methods (e.g. chemistry, biology, pharmacology and molecular imaging). The specific research contents are as follows: (1) Developing visualized drug delivery systems based on in-vivo optical imaging (such as Raman imaging) or nuclear medical imaging technologies, for intra-operative diagnosis and removal of residual micro-tumors, reversing targeted-drug resistance, medication guidance and treatment evaluation; (2) Investigating oral drug delivery system for naked-eye visualization and treatment of colon diseases at the early stage; (3) Designing DNA-modularied programmable bivalent drugs for precise receptor regulation. Recently, we have published more than 40 research articles in the international academic journals (e.g. Chem Soc. Rev., Nature Comm. Angew. Chem., Nano Lett, ACS Nano, Adv Funct Mater., etc), and three Book Chapters. Among them, there has been 1 article cited more than 1000 times, 5 articles cited more than 200 times, 7 articles cited more than 100 times, and 3 articles selected as the highly cited papers by ESI. One visualized drug delivery system has begun clinical trial stage. In addition, we also have applied/authorized two international patents and one domestic patent.

Lab Info:Our laboratory aims to integrate molecular design with delivery strategies to engineer innovative therapeutics and tools addressing current clinical challenges. With a focus on the targeted protein degradation field, we employ an interdisciplinary approach that combines chemical biology, medicinal chemistry, drug delivery, and biomedical engineering to advance research across three interconnected themes: (1) designing and synthesizing functional molecules; (2) elucidating their mechanisms of action under disease conditions; (3) incorporating them into advanced delivery systems for therapeutic applications. Research from our group has been published in high-impact journals, including Nature Biotechnology, Nature Reviews Clinical Oncology, Journal of the American Chemical Society (JACS), and Chemical Society Reviews. We have also filed multiple PCT international patents, U.S. patents, and Chinese invention patents.
