The sequencing core provides a set of services from high-throughput next generation sequencing (NGS) to downstream bioinformatics analysis. The main services include NGS experiment design, nucleic acid samples quality control, sequencing library preparation, on-machine sequencing and subsequent gene annotation and following data analysis.

The platform is fully equipped and capable of nucleic acid process; The platform is equipped with Covaris M220, BluePippin (automatic nucleic acid electrophoresis and fragment recovery system), and Agilent 2100 bioanalyzer. The core has two sequencers, Illumina NextSeq500 and Illumina MiSeq, three server, Sugon, Inspur and Fenghu, for sequencing and data analysis. The core has built three single-cell sequencing platforms, Fluidigm C1, 10x Genomics and Drop-seq, which fully meet various single-cell sequencing needs; Nanopore sequencer serves third-generation sequencing; IsoLight single-cell protein analyzer is used for single cell cytokine detection.

The platform provides technical services such as single-cell sequencing, whole-genome sequencing, whole-transcriptome sequencing, metagenomic sequencing, exome sequencing, 16S bacterial population sequencing, small RNA sequencing, and long-chain non-coding RNA sequencing. It also has data processing pipelines for downstream biological informatic analysis.

The platform provided mature and diverse single-cell sequencing technology. The 10x Genomics single-cell platform provide technologies include 3'-end transcriptome sequencing, 5'-end transcriptome sequencing, VDJ immune repertoire sequencing, and single-cell ATAC-seq. Technologies that mixing multiple samples and overloading, and capture membrane proteomics together with single-cell transcriptome has been realized (Cite-seq, Total-seq). Moreover, the platform is equipped with BD’s Rhapsody single-cell analysis system demo machine.

The diagnostic sequencing core serves Shanghai Immunology Institute, Shanghai Jiaotong University School of Medicine and other academic institutions. Besides, the core also cooperates with Meiji Biology company and Shanghai Daan Medical company.
