Abstract :

It emerges that the ribosome, through the diversity of its composition, is a direct player in translational control. Our team has shown for the first time that ribosome composition is chemically modified during tumorigenesis (Marcel et al, Cancer Cell 2013; Erales et al, PNAS 2017). Alterations in ribosome biogenesis have also been linked to the activation of epithelial-mesenchymal transition (EMT) in cancer cells (Morin et al., Biochim Biophys Acta Rev Cancer 2022). In particular, up-regulation of the 57 kD intermediate filament protein, Vimentin, in mesenchymal cancer cells, is correlated to increased ribosome biogenesis (Prakash et al Nat Com 2019). The PhD project presented here aims at understanding the alterations in ribosome composition occurring in human metastatic colorectal cancer cells (CRC) that have undergone EMT. The main focus will be the elucidation of the translational and spatial regulations of vimentin and other EMT factors induced in CRC cell lines with altered levels of ribosome biogenesis. The perspective of this PhD thesis project is to identify key factors involved in ribosome biogenesis regulation of vimentin and EMT factors expression in order to find new therapeutic targets to treat metastatic colorectal cancer.
